The Mechanism of the Secretion of Sulfonamide Drugs in Gastric Juice *
نویسنده
چکیده
Cooke, Davenport, and Goodman4 have shown that five sulfonamide drugs are secreted in the gastric juice. When the concentration ratio is defined as the concentration in the gastric juice divided by the concentration in the plasma it is found that the average concentration ratios of sulfanilamide and sulfapyridine are 2.6 and 3.2 respectively. The average concentration ratios of sulfathiazole, sulfadiazine, and acetylsulfanilamide are 0.16, 0.28, and 0.30 respectively. The concentration ratios of all the drugs decrease significantly with increasing rates of gastric secretion, and the concentration ratios of all but sulfathiazole increase significantly with increasing plasma concentration of the drugs. The fact that the concentration ratios of sulfanilamide and sulfapyridine are greater than one makes it appear that osmotic work is done by the gastric mucosa in secreting the drugs. This cannot be explained by the tentative theory of the mechanism of acid secretion proposed by Davenport' or by any other current theory. The meaning of the variations of the concentration ratios with rate of secretion and plasma concentration is also obscure, and it must be explained before any theory of the mechanism of secretion can be accepted. This paper presents a simple and comprehensive explanation of all the observed facts of sulfonamide secretion, and it shows that the facts are not opposed to Davenport's theory of the mechanism of acid secretion. The equation for evaluating the work done when N mols of a solute are transferred from one solution where its activity is fp [PI to another in which its activity is fgj [GJ] can be written

The Secretion of Sulfonamide Drugs in Gastric Juice *
Although the sulfonamide drugs are ubiquitously distributed throughout the entire body water, a property important for their wide chemotherapeutic value, quantitative differences do exist with regard to the concentrations attained in certain compartments of the extracellular fluid after equilibrium is established. Perhaps most striking is the failure of sulfathiazole to reach a concentration in...
متن کاملEffect of esophageal distension on gastric secretions in rat and the possible mechanism
Introduction: The effect of esophageal distension (ED) on gastric motility has been well documented, but a few investigations have been carried out on the effect of ED on gastric secretion. The aim of this study was to investigate the effect of ED on gastric acid and pepsin secretion and the mechanism(s) involved. Methods: Male adult Wistar rats (200-240 g) were anesthetized with urethane (1...
متن کاملComparison of the Effect of Cydonia oblonga and Phyllanthus Emblica on Gastric Residual Volume and Pulmonary Aspiration in Patients under Mechanical Ventilation in Mousavi Hospital ICU of Zanjan in 2020
Background and Objective: The gastric residual volume has a direct effect on the volume and timing of the subsequent feeding and causes gastric emptying and pulmonary aspiration. The present study compares the effect of Cydonia oblonga and Phyllanthus Emblica (Amla) fruit on gastric residual volume and pulmonary aspiration in mechanically- ventilated patients. Materials and Methods: This is a...

متن کاملThe role of D1/DA1 dopamine receptors on histamine-and carbachol-induced gastric acid secretion in male rat
Dopamine has an effective role on gastric acid secretion (GAS). The role of peripheral and central dopamine D1/DA1 receptors on stimulated GAS are unknown. The objective of the present study was to use SKF38393 and SCH23390 on stimulated GAS. For all gastric sampling, animals (Sprague Dawley rats) were anesthetized and a polyethylene tube was introduced into the stomach through esophagus and a ...
